Determination of the microbiological activity in biogas plants

In the field of biogas production a lot of new innovative technologies were developed in the recent years. However, none of these technologies reveals or influences the workflows and activities of the microorganisms in the biogas process. But only a sound microbiology guarantees a successful biogas production.

The aim of this work is to develop a method to determine the activity of the methane-producing microorganisms in biogas plants through molecular biologic techniques in a quantitative way. By this, the influence of common fermentation parameters like pH, temperature or substrate composition on the microbial community can be studied and specifically modified.

The possibility to measure and influence the microbial actions in the process of anaerobic digestion leads to an increase of the plant efficiency and a better utilization of renewable resources. Thus, this work makes a substantial contribution to the technological progress and the use of biogas as renewable energy source.

The project contains the following tasks:
  1. Development of the molecular biological method for measuring the microbial activity in biogas plants:

    • Extraction of mRNA from biogas plant fermentation samples
    • Reverse transcription (RT) of the extracted mRNA into cDNA (complementary DNA) as well as amplification, detection und quantification of the cDNA by means of RT-qPCR
    • Validation of the method
    • Evaluation of the method by means of 16S rDNA and mcrA clone libraries (TRFLP)
  2. Application study of the new developped analysis method: Using the new set-up in order to test the effect of micro-nutrients of the partner company on the bacterial activity
